1. Nucleoli in nuclei of a Vicia faba root tip cell (except lower right, wich is Arabidopsis).
Top left - unstained, uncontrasted;
Top centre - DIC contrast optics, indicating the presence of a marked change in refractive index at the boundary of the nucleolus;
Top right - iodine staining, indicating dense content of protein and nucleic acid in the nucleolus;
Lower left - toluidine blue staining, purple metachromasy indicating presence of acidic material (RNA, not DNA) in the nucleolus;
Lower centre - Fuelgen reaction, confirming low content of DNA in the nucleolus, relative to the stained chromatin elsewhere in the nucleoplasm;
Lower right - silver staining, a general stain for nucleoli.
